I am a permanent resident of US. (Green Card holder) Can I work in Canada ? Does US green card give a right to work in Canada ?

Permanent residence in the USA has no significance for Canada except to assure that you are likely to return to the USA after a visit, and so make obtaining a visa (if you need one) easier.
Nor is it relevant to working in Canada, and it doesn't afford NAFTA rights. On the other hand, you should be able to travel to Canada on behalf of your US employer on, say, a business or sales trip.
Technically, you have no rights even with respect to cross-border factories (of which there are a few). But sometimes technical rules are ignored: imagine the staff of the Haskell Free Library and Opera House being told they could not perform any function in the half of the building that is in the "other" country than that of their nationality.

It's nearly impossible for a US citizen to work in Canada, never mind a green-card holder. The only way I know of and the most common is company sponsorship.
Of course this requires the company to essentially say, "Hey I really like you! In fact I'm going to tell my government to allow you to come over here to work for me instead of hiring Joe Smith who lives 2 blocks away, won't cost me any extra money and won't require 5,000 miles worth of beurocratic red-tape."
