She can't do without your permmision, you have the controlling vote in this matter. She could do some major psychological damage to your child, which is abuse, which makes her a perpetrator against minors. Get her some counselling, get some legal advice for yourself, or ask her to leave. There is something wrong with your wife. Because there are people out there who can not have kids of their own and would give anything to have their own kid regardless of the gender of the child. She needs to realize that.She should be grateful her child is healthy. I know I am grateful for my children.

Your son will have already realised that mummy doesn't like him much and that he is a disappointment, you have a duty to repair the damage already done and to raise your son so he has a sense of self worth. Even tho your son is young he has already realised he isn't good enough for mummy so for gods sake put that little boy first, he is the most important person in all of this not you and not your wife who needs some sort of counselling to help her change her outlook on life and your son.

DO NOT have any more kids with your wife, she is not psychologically capable of raising a baby no matter if it's a boy or girl as I think she has a fantasy idea of life with a baby girl & real life will ruin that.
You cannot trust her to not harm the baby in some way because her fantasy has not come true.

just the fact that you asked this question tells me that your wife has serious issues. Before she decides to go ahead with this i think you should try to get some help for her as soon as possible. I think she is suffering from post partum depression and is unable to think logically. It is therefore your responsibility to protect your son and get some help for your wife. Keep him away from her until she gets the proper help if necessary. You have to find a way to get her some help so that she can begin to think logically before this happens.
Post partum affects women in different ways and they are unable to see anything wrong with their train of thought. Please find a way to get help for her and get her family and friends in on it to convince her that she needs help.

if she cant love a child that she not only carried for nine months, but she also took care of him for two years...you need to step up, because she has problems...does she really even love you...i know that child loves the two of you...so if either of you can turn off love like that for him you need to seek help, because there is something seriously wrong...i love the h-e-l-l out of my children and i have from day one...i could never just give them up like that...and i have teenagers that never stop getting in trouble...keep your child, love your child, and never let him go...how would you feel if she gave you up after two years...well that child will feel a million times worse because he was give up because he was a boy....what happens if you have a girl and she grows up to be gay...

Take your son and get rid of your wife. A child needs his parents and it seems she would not be a good one for him. Raise him by yourself and you will have created a better adult in the long run. Good luck.
