The attack on Ft Sumter. And, there was nothing civil about it, either call it the War of Northern Aggression or the War Between the States.

Sumter was the first attack (naval), and the Battle of Philippi Races was the first organized land action, even though it is dismissed by some as a skirmish between Union and Confederate soldiers. Manassas (Bull Run) was the first major land battle to take place.

Actually, the first battle(verbal) was between the Constitutional Delegates of the 13 States when they were framing the Constitution. Bitter exchanges between the slaveholding delegates and the others led nowhere and the first compromise to appease the slaveowners started the United States toward the bitter disintegration of the Union between 1861 and 1865.

Abiding by your hint, the first battle(physical) was John Brown and his ragtag band against the Virginia militia under Robert E. Lee. This small demonstration of the fanaticism of Abolitionists gelled the resolve of slaveowners and their "pocket politicians" to brook no further interference by either Abolitionists or their politicians.

The first shots fired by a seceded state, South Carolina, were on January 9, 1861, when artillery forced the Star of the North to retreat from its attempt to resupply Fort Sumter.

The first exchange between forces of the respective Nations was at Fort Sumter, April 12, 1861.
